In dairy farming, breeding bull subfertility is a well-known issue. Thousands of female cows are bred from the semen of a single bull. Evaluation of the quality of the semen plays a major part in fertilisation. Sperm RNA transcripts are essential for spermatogenesis, sperm maturation, fertilisation, activation of the oocyte genome, embryogenesis, and placental development, as well as for capacitation, motility, and fertilisation conditions. Establishing RNA isolation from sperm HF bull using various techniques was the goal of this study. Four different methods are used for the isolation of RNA. RNA yield in bull spermatozoa of the combined method resulted in the highest. Origin Total RNA Isolation Kit, Double TRIzol method, TRIzol method showed the results decreased RNA quality receptively.
